Immunochemical Identification of the Main Cell Wall Polysaccharides of the Early Land Plant Marchantia polymorpha

Plant primary cell walls are composite structures surrounding the protoplast and containing pectins, hemicelluloses, cellulose polysaccharides, as well proteins. Their composition changed during evolution of green lineage from algae to terrestrial plants, i.e., an aquatic a environment. The constraints life in environments have generated new requirements for organisms, necessitating adaptations, such wall modifications. We studied polysaccharide thalli Marchantia polymorpha, bryophyte belonging one first land plant genera. Using collection specific antibodies raised against different epitopes, we were able identify polysaccharide-enriched fractions: including low-methylesterified homogalacturonans; rhamnogalacturonan I with arabinan side-chains; xyloglucans XXLG XXXG modules, mannans, galactomannans, xylans. could also show even distribution galactomannans by immunocytochemistry. These results discussed regard proteome context lineage. polysaccharides M. polymorpha illustrate transition charophyte ancestors plants xyloglucans, xylans mannans embryophytes which do not exhibit major polysaccharides.
